Far as I know, this is an AMD only thing.
I think it's Phenom II and Athlon IIs as the only ones that have potential to unlock. Even if you do unlock other dormant cores, there's a high likelihood for instability in your system. These chips got binned down for some reason (some say to meet demand for lower price points, but I'm not convinced).
I had three Phenom II 555's and two unlocked- one was a tri-core and the other was a full quad, but didn't overclock well. The last one didn't unlock at all. Luck of the draw.
The motherboard plays an important role here as well. Double check to make sure that the chipset is 880 or newer.
For most people this is a nice treat, but if you're looking to get the best of the best, then just buy what you want.
In the first iterations of the Phenom II, you could unlock the 4th core in the X3 720 and some others.
Since then, I believe most of them are laser-etched out. Unlock with care.
